Denture Alternatives on the Horizon: Japan Explores Tooth Regeneration

It’s well known that smiling boosts the secretion of arousal hormones while lowering levels of stress hormones like cortisol and adrenaline.

However, for some people, smiling can bring hesitation—either because they don’t want to show missing or damaged teeth or because they worry about the appearance of dentures.

While traditional dentures can be helpful, researchers in Japan are investigating a groundbreaking alternative: using drugs to regenerate natural teeth.

Promising Laboratory Trials

The research, led by Dr. Masaru Takahashi of Kitano Hospital’s Graduate School of Medical Research, aims to develop a drug that could stimulate the growth of new teeth by 2030.

Early experiments target the protein USAG-1, which is known to inhibit tooth development. Blocking this protein in mouse models can trigger the growth of extra teeth.

The team also discovered a “third set” of teeth in the human mouth, present as dormant buds. While supernumerary teeth (extra teeth in addition to normal teeth) occur in only about 1-3% of the population, this discovery suggests that drugs could activate these hidden buds and allow them to grow into fully formed teeth. Unlike primary and permanent teeth, this third set of teeth represents an untapped natural regenerative resource.

The Future of Oral Health

A global survey covering 19 countries found that nearly 19% of people wear some form of denture. While dentures can restore function and aesthetics, they typically require a period of adjustment, can be uncomfortable, and can slip while speaking or eating. They also require rigorous cleaning to prevent bacterial growth and further oral health problems.

The possibility of regenerating permanent teeth represents a revolutionary advance in dentistry. It offers new hope to people who have lost teeth due to injury, disease, or genetics. With continued successful trials, the future may not only be filled with brighter smiles, but also more natural ones.
